The annual wild weed species Arabidopsis thaliana is a model organism not only for molecular biology but also for ecological and evolutionary genetics, and hence, revealing the geographical structure of its genetic variation has become of paramount relevance.
Furthermore, identification of genetic structure is relevant to mapping the causal genes responsible for the natural variation of adaptive traits by genomewide association analysis.
The genome of the flowering plant Arabidopsis thaliana has five chromosomes
the largest, chromosome 1, in two contigs of around 14.2 and 14.6 megabases
Chromosome 1 contains about 300 gene families with clustered duplications

Salt Gradient / Halotropism

Preparing Salt Gradient

Sterilise glass cutting tool in alcohol (70% EtOH)

Take out glass cutter and let dry up

One might dry up EtOH with flame, however be careful not to break the glass

Cut the diagonal and scoop out the 0mM agar

Throw left over 0mM (control) agar in a bag (prepare to have installed ;)

Make sure to carve a diagonal & neat, straight line

Results half way. Remember to make some extra plates

Next step: add 200mM Salt (NaCl) in the carved out triangle

Now this is quite a precise job as we don’t want elevations in the changeover

Result: Salt Gradient 0mM to 200mM NaCl

Next time we’ll do the diagonal the other way around
